Physical form
The product is a recombinant Shiga toxin 1 B subunit, which contains 71 amino acid residues and an HIS tag at
the C-terminus. It is frozen in DPBS solution without any carrier protein.
Analysis Note
Gb3-binding activity: significant binding above the background is achieved with ≤60 ng/mL of STxB. The activity
of STxB is measured by its ability to bind to Gb3, which requires its pentameric form.

H1299 was incubated with STxB (AF488) for 40 minutes at 37°C to observe binding to the plasma membrane
and intracellular uptake. The experiment was performed without preincubation of STxB with H1299 at 4°C. The
cells were washed twice, fixed, permeabilized and stained with antibodies to visualize the trans-Golgi
compartment (TGN46). Nuclei were stained with DAPI. Scale bars represent 10 µm. Clear STxB accumulation
intracellularly after 40 minutes of incubation without pre-incubation treatment.
